Flow cytometry sheath fluid is the solution that directs the alignment of cells into a single-file stream under controlled conditions as they pass through a flow cytometer. The sheath fluid that is most suitable varies on experimental needs and each formula has its advantages. Sheath fluid formulations commonly include phosphate-buffered saline (PBS) and HEPES-buffered saline. HEPES-buffered saline controls pH better at high pressure than phosphate buffers and is ideal for high-pressure cell sorting. Solubility, pH, osmolarity, and compatibility with the instrument are important factors to consider when selecting sheath fluid.
