Klenow Fragment

The Klenow fragment is a truncated version of the E. coli DNA Polymerase I that has lost the 5’-3’ exonuclease activity while retaining the original 5'-3' polymerization and 3’-5’ exonuclease and proofreading activity. The fragment arises from cleavage of the polymerase with the serine endopeptidase subtilisin. The Klenow fragment is useful for performing various DNA modifications, including DNA synthesis from single-stranded templates, DNA blunting of overhangs, creating DNA probes, dideoxy sequencing, and DNA labeling.
