For quantitative detection of human FABP2 in cell culture supernates, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).
Background: Fatty acid-binding protein 2 (FABP2), also known as Intestinal-type fatty acid-binding protein (I-FABP) is a protein that in humans is encoded by the FABP2 gene. The intracellular fatty acid-binding proteins (FABPs) belong to a multigene family with nearly twenty identified members. FABPs are divided into at least three distinct types, namely the hepatic-, intestinal- and cardiac-type. Using a human cDNA probe for in situ hybridization studies, they regionalized the assignment to 4q28-q31. The intestinal form of FABP was mapped to mouse chromosome 3 between the amylase and alcohol dehydrogenase-3 loci. Also, they may be responsible in the modulation of cell growth and proliferation.
Principle: Human FABP2 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from Mouse specific for FABP2 has been precoated onto 96-well plates. Standards (Expression system for standard: E Coli, Immunogen sequence: A2-D132) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for FABP2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substRate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution.
