Clone Number:

UCHT1

This antibody has been used to optimize flow cytometry-based immunophenotyping of PBMC samples. The results have not yet been published. Multi-colour antibody panels were designed to identify several lymphocyte populations and tested to ensure appropriate spectral unmixing was possible in a 20+ color panel.

Experimental Design and Results Summary

Applications

Flow cytometry

Sample

Human PBMC

Primary Incubation

5µL Spark Violet™ 538 CD3 for 30 minutes at 4°C

Blocking Agent

Human TruStain FcX™ (Biolegend 422302) and CellBlox™ Plus (Thermo Fisher C001T06F01)

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

3L Cytek Aurora, Kaluza Software

Results Summary

Staining allowed the distinction of CD3+ T cells. Titration of the antibody was conducted, although this resulted in loss of separation between positive and negative populations. This antibody fit well into a 20+ colour panel with no unmixing issues.

Summary

The Good

Distinct staining allowing selection of T cells.

The Bad

Titration resulted in loss of distinct populations.

The Bottom Line
