The PE Cy5 CD64 antibody works well for confirming the high expression of Fcgamma receptor I on alveolar macrophages, absent on DCs, within immune cell populations isolated from mouse lungs and bronchoalveolar lavage fluid. The antibody is also quite cheap.

Results Summary

I used this product confirm that my gated population was alveolar macrophages, in mice treated with PBS or clodronate liposomes. Cells were isolated from BALF using 1 ml of 1% BSA in PBS and from lungs through collagenase digestion; RBC lysis was performed using ACK buffer. Cells were then stained with viability dye followed by extracellular antibodies such as CD11b, CD11c, Siglec-F, CD64. The alveolar macrophage pool looked very clear from the live cells. Using CD11c and CD11b, alveolar macrophages were distinguished from eosinophils. CD64 expression on these cells was then confirmed as shown in the picture. I have not seen any spillover issues with this antibody and fluorophore combination, despite alveolar macrophages being auto fluorescent.

The Bottom Line

I would highly recommend this antibody for myeloid cell identification of macrophages versus dendritic cells. The fluorophore is compatible with PE cy5.5 as well as PE, yet it is a common fluorophore so I would recommend designing panels well in advance before purchasing this antibody.
