In order to maximize the transduction efficiency of my primary T cells, I need my T cells to be sufficiently activated. I use this CD3 OKT3 antibody to activate my T cells before transduction and have consistently had high-quality activation.
T cell activation
Primary human T cells
Prepare antibody into 1 ug/mL into DI water and incubate plate overnight at 4 degrees C.
Remove solution, and wash twice with media before plating T cells.
Plate T cells on antibody-coated plate overnight in the incubator

This product along with anti-CD28 mAb helped activate and expand/proliferate my human T cells before lentiviral transduction. CD3/CD28 activation is pertinent for high efficiency transduction and helps to promote high T cell expansion of both CD4 and CD8 subsets.
