Proteins resolved in 10% SDS-PAGE were western transferred in a nitrocellulose membrane and probed for the Hexokinase II protein. This experiment aimed to test the time course effect of a specific treatment on Hexokinase II expression in a human primary cell line. The selection of antibodies from Abcam Systems was due to the excellent band of Hexokinase II shown in the antibody flyer.
Western blot
Human whole-cell protein lysate.
Dilution (1:1000) in 5% BSA prepared in 1x TBST, overnight incubation at 4 degrees celsius.
5% skimmed milk prepared in 1x TBST.
Goat anti-rabbit IgG HRP Ab, Dilution (1:4000) in 5% skimmed milk prepared in 1x TBST, 1hr at RT.

Enhanced chemiluminescent detection in X-Ray films.
In the present study, Hexokinase II antibody from Abcam was used. It was a nice feeling because this antibody has shown only a specific and bright band of Hexokinase II which showed an excellent specificity of this antibody for Hexokinase II epitope.
Detected highly specific band of Hexokinase II. Got not a single non-specific band.
