The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. The goal of this experiment is to determine expression levels of Cas9 fusion proteins for future CRISPRi experiments.

Results Summary

KRAB-dCas9 and dCas9-KRAB-MeCP2 were stably expressed in HEK293T cells. 50 µg of whole cell extracts were resolved on a 4-12% Bis-Tris gel and proteins were transferred onto a nitrocellulose membrane. This antibody detects stably expressed Cas9 fusion proteins.
