The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. The goal of this experiment is to determine if this antibody works for immunoprecipitation of endogenous APC.
Immunoprecipitation
RKO whole cell extract
5 mg of whole cell extract was incubated with 2 µg of antibody for 16 h at 4 deg C on a rotator.
5% milk in TBS-T for Western Blotting step.
10 µL of Protein A Sepharose magnetic beads for Immunoprecipitation step.

Immunoprecipitate complexes were eluted from the magnetic beads and resolved on 3-8% Tris Acetate gel followed by Western blotting with a different APC antibody.
Whole cell extracts from RKO wild type (APC full length) and RKO APC truncated cell lines were used in this experiment. This antibody is really good for immunoprecipitation and works well when incubated for 16-20 h at 4 deg C.
