Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of antibody secreting cells (ASC) and B cells in health and disease. Here, we are just titrating an anti-TIGIT antibody for posterior use.

Results Summary

To titrate the antibody we used spleen cells and gated in CD4+ T cells, since some are suppose to express some TIGIT. We tested several dilutions and 1/100 seems to be the one giving better results. As it can be seen in the figure, we can detect around 3,6% of CD4 cells expressing TIGIT.
