We regularly detect apoptosis in several human cancer cell lines using the Caspase-Glo® 3/7 Assay from Promega. The protocol is easy to handle and the measured luminescence is robust, reliable and very sensitive (even with small amounts of cell suspension).

Starting Material

Human cells in a white-bottom 96-well plate, conventional luminometer.

Tips

We obtained good results even with 10 % of the stated glo substrate volume. Just make sure the ratio of cell medium and substrate remains 1:1 (e.g. 10 µl cells + 10 µl substrate). Can easily be combined with generation of Western Blot samples.

Results Summary

We obtained robust cell death data for several treatments/conditions in human cancer cell lines e.g. in a recent project about the synergism of the BH-3 mimetic ABT-199 and proteasome inhibitor Bortezomib.
