One week after depleting Natural Killer cells from a B6 mouse, we used the FITC anti-NKp46 antibody to analyze the lymphocytes in the spleen using flow cytometry to detect the presence or absence of Natural Killer cells.

Results Summary

The FACS analysis using the FITC anti-NKp46 antibody worked excellently when used in combination with a BV605 anti-NK1.1 antibody. When we compared the splenic lymphocytes of a control mouse vs a mouse that had received an anti-NK1.1 treatment, we saw little to no Natural killer cells in the treated mouse. The FITC anti-NKp46 antibody helped us to successfully identify the presence or absence of Natural killer cells in our treated vs control mouse.

Summary

The Good

The results were specific and reproducible. It did not create compensation problems when used with 10 additional fluorophores.

The Bad

We had to use a significant amount of antibody.

The Bottom Line

Although we had to use a significant amount of antibody, it was very efficient in labeling low and high expressers of NKp46 and helped us specifically identify NK cells. I continue to use this antibody for all of my FACS analysis of Natural Killer cells.
