We work on kidney and renal podocytes development. The podocytes marker Nephrin (NPHS1) is crucial to identify and follow the maturation and functionality of this cell population. This antibody showed very good staining in different conditions and tissue origins.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Mouse renal samples

Primary Incubation

1:200 in blocking solution, 1 hour at RT

Blocking Agent

PBS + donkey normal serum 10%

Secondary Incubation

Fluorescence coupled secondary antibody, 1:500 for 1 hour at RT

Tertiary Incubation

None

Detection

Fluorescence/confocal microscopy

Results Summary

Very strong and specific staining in mouse kidney slides, even using secondary antibodies know to have high background in this tissue (ie Alexa488 or Alexa555). It works in frozen tissue (OCT embedded) as well in paraffine embedded samples. It stands all the unmasking condition and pH we tested so far.
