About 4 million thymocytes isolated from a 5-7 week old C57BL/6 mouse were stained with 5 different antibodies, including BV605 conjugated anti-mouse CD24. 0.1ug (which is 0.5 uL in volume) of CD24-BV605 was used to stain the thymocytes, which resulted in a very bright staining signal. The CD24-negative and -positive peaks were well resolved. Compensation with the conjugates APC, PE, FITC and APC/Cy7 was fine.

Results Summary

The CD24 non-expressing cells were well resolved from the positive population. For thymocytes, all the developing CD4+CD8+ (double positive) T cells express high amounts of CD24. The maturing CD4+ or the CD8+ (single positive) cells downregulate CD24, with the more mature cells having reduced expression of CD24.
