In our lab, we are purifying neutrophil granulocytes from human peripheral blood for down-stream analyses. A central step during this process is the routine assessment of purity, which we determine by staining for CD16 and CD15. Both markers are highly expressed on mature, peripheral blood neutrophils in humans. The anti-human CD16 antibody is part of our routine assessment panel and has yielded nothing but exceptional results.

Results Summary

According to manufacturer's instructions, we stained the neutrophils in a 1:50 dilution of CD16 FITC antibody. The antibody neither required titration nor buffer optimisation or Fc Blocking, allowing us to screen neutrophil purity in a fast and reproducible way. Additionally, the isotype control was negative. Even though, the supplier changed the formulation of the product during our studies, we could use the new dilution as provided and it still worked well with our established pipeline!
