To test the time course effect of particular growth factor on cyclin E2 expression in mouse, the primary cell lines were growth-arrested for 12 hrs in medium devoid of FBS. The synchronised cells treated without or with growth factor for an indicated periods were employed to prepare proteins extracts, and an equal amount of proteins were resolved in 10%SDS-PAGE. The resolved proteins were then western transferred in a nitrocellulose membrane and probed with cyclin E2 specific antibody. Although, Thermo Fisher Scientific supply a polyclonal antibody, the selection of Ab was based on the merits indicated in antibody flyer.

Results Summary

Although Cyclin E2 from Thermo Fisher Scientific was a polyclonal antibody, it has detected only a specific band at the correct molecular weight (compared with low range protein marker, Bio-Rad# 161-0304). Interestingly, a highly specific band with high Intensity was observed upon growth factor treatment.
