MC38 tumors were excised from B6 mice and digested with Collagenase IV (1 mg/ml) and DNaseI (200 ug/ml). Cells were then stained with L/D Blue for gating out dead cells. Subsequently, cells were stained with a range of different antibodies to identify a range of immune cell types. CD4- fitc from Biolegend was one of the antibodies used at a 1:200 dilution. Antibodies were incubated with the tumor cell suspension for 30 minutes at 4 deg C.

Results Summary

CD4+ cells could be clearly gated after tumor cell suspension was stained with antibodies to identify immune cells. First, cells were gated on CD45, then on TCRb and then output for CD4- fitc (1:200).
