We transformed 5ul of ligation mix into one shot stbl3, kept on ice for 30 min and heat shocked for 45 sec at 42 c. Added SOC medium provided with the kit and incubated for 1 hour at 37c. Plated 10 ul of SOC medium on to LB/AMP plates and incubated O/N at 37c. Observed well-spaced single colonies on all the transfected plates.
