Phospho-PAK1 (Ser199/204)/PAK2 (Ser192/197) Antibody

Catalog Number:

2605

In the present study, pPAK1 + pPAK2 levels were measured in the muscle of rats subjected to ischemia. For this, male albino Wistar rats' femoral arteries were ligated and cut, and after 7 days of recovery, femoral muscles were harvested. Proteins were extracted from samples resolved in SDS-PAGE and immunoblotted for phospho-PAK1/PAK2 levels. The antibody was selected based on previous experience of the same antibody with human samples, and it works well in rat, too.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Whole tissue protein lysate

Primary Incubation

Overnight at 4°C (5% BSA in TBS with 0.1% Tween 20; dilution 1:1500)

Blocking Agent

5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)

Secondary Incubation

2 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

Detection

ECL

Results Summary

The cell signalling phospho-PAK1/PAK2 antibody has given a very specific band of pPak 2 and Pak1/3 at the correct size. However, there was a significant change in pPak2 levels in response to treatments.

Summary

The Good

Antibody is highly specific to phospho-PAK1/PAK2

The Bad

A nonspecific band was also observed at 26 kDa

The Bottom Line
