High-quality genomic DNA extraction from mammalian cells is a common step in most molecular biology applications such as PCR, qPCR, and NGS. Here, I utilize the QIAamp DNA kit to extract gDNA from the A549 lung cancer cell line.

Protocol Overview

This kit allows cell pellets to be resuspended in a Proteinase K lysis buffer that allows efficient extraction of gDNA. The samples are kept in a 56°C water bath for 10 mins and then placed on a column for downstream purification. You can also treat the sample with RNAse A to eliminate any RNA contaminants in your prep. This is highly recommended as the 260/280 ratio of your samples will be >1.8 if you just run your sample using this kit.

Set up a water bath in advance and treat the samples with RNAse A to obtain purified gDNA

Results Summary

This kit is fairly robust and can be multiplexed. Ideal for experiments that require gDNA extraction for downstream NGS applications (Chip–Seq, etc.). The samples were treated with RNAse A after purification to eliminate any RNA contamination. Samples were utilized for PCR.
