I am interested in studying the molecular mechanism of hedgehog signal transduction in vertebrate cells. I had performed a CRISPR/Cas9 genetic screen to uncover new regulators of hedgehog signaling pathway. One of the negative regulators identified in these screens is known to regulate a cell surface receptor relevant to hedgehog pathway. The goal of this experiment is to determine if this gene affects the overall levels of one of the WNT receptors, FZD3.

Experimental Design and Results Summary

Applications

Western Blot

Sample

NIH-3T3 whole-cell extract

Primary Incubation

1:200 dilution in 1xTBS-T; overnight at 4°C for 16 hr

Blocking Agent

5% milk in TBS-T

Secondary Incubation

1:10,000 dilution of donkey anti-mouse IgG+HRP; 1 hr at room temperature

Tertiary Incubation

None

Detection

ECL pico plus

Results Summary

This antibody detects a band at ~85 kDa. The predicted size is 76 kDa. Signal is weak and requires ECL pico plus and long exposure.

Summary

The Good

OK for Western Blotting

The Bad

Weak signal

The Bottom Line

Weak signal with this antibody in NIH-3T3 cells. Signal-to-noise ratio can be improved by loading more protein.
