CD56 is a general marker for natural killer (NK) cells and is also expressed on a subset of CD3+ T cells. One of the focuses of our lab is studying the NK cell and NK-T cell response in the context of viral infection. In summary - a good antibody for identification of CD56+ cells.

Human PBMC isolated from whole blood by Ficoll

Primary Incubation

FITC-conjugated anti-human CD56 antibody, incubated with approximately 1x10^6 cells for 1 hour on ice at 1:5 dilution (20ul) in a total volume of 100ul PBS+2% FCS, 1mM EDTA, 0.1% sodium azide.

Results Summary

CD56+ cells were relatively easily distinguished from CD56- cells; however, they were not that bright. Other antibodies attached to brighter fluorophores allow clear resolution of CD56+bright and CD56+dim NK cell populations. It is difficult to identify these populations with this antibody.

The Bottom Line

A good antibody for identification of CD56+ cells. If you are interested in distinguishing between CD56+bright and dim populations, use a different clone directly conjugated with a brighter fluorophore.
