I am staining gamma delta T cells in mouse lungs. Our goal is to clearly separate them from TCR beta positive and CD1d tetramer positive T cells. I tested multiple different antibodies and fluorophores, and found this to be one of the best.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Collagenase-digested mouse lung tissue

Primary Incubation

1:200 for 20 minutes on ice

Blocking Agent

24G2 in 3% BSA in PBS for 10 minutes on ice

Secondary Incubation

Fixed with 4% formaldehyde for 30 minutes

Tertiary Incubation

none

Detection

Flow cytometer

Results Summary

Our FACS results showed one clear CD45+ gamma delta TCR+ population in the lung sample. There was no detectable staining on other T cell subsets, or any other immune cells. There was no detectable TCRgd high and TCRgd low population, as some antibodies show.

Summary

The Good

Stains specifically for gamma delta T cells, with no detectable non-specific staining

The Bad

The fluorophore is relatively dim compared to others

The Bottom Line

If your bright color channels are already used by dimmer markers, this is a very suitable antigen for labeling with PerCP-Cy5.5. The staining is clear, and good separation is highly achievable. Highly recommend.
