Custom peptide

The peptide was ordered to bind specifically to receptors. It was used to show the suppression of GPCR receptors on CaCO2 cells. When this peptide was added at the concentration of 0.2 mg/ml in confluent cells and incubated at 37 oC for 24 hrs, the expression of GPCR was found to reduced drastically. While the peptides obtained from other sources needs larger dose (< 0.2 mg/ml) this peptide works at much lower dose.

Results Summary

The costume peptide was obtained on 5th day of ordering in the form of lyophallized vial. It was then reconstituted in PBS pH 7.4 at the concentration of 2 mg/ml and frozen until further use. Cells were grown in 6 well pate until they reach confluence. Cell medium was removed and washed twice with PBS. Followed by this diluted peptide at the concentration of 0.2 mg/ml with serum free F12 medium was added to the cells. Each well contains 2 ml of the peptide solution. Cells were then incubated at 37 oC for further growth. After 24 hrs cells were taken out, medium was removed and cells were lased using trypsin. Expression of GPCR in cell lysate was carried out using real time PCR and correlated to that of untreated.
