Invitrogen Antibodies

Product Name:

CD127 Monoclonal Antibody (A7R34), APC

Catalog Number:

17-1271-82

We are an immunology lab studying CD8 T cell memory differentiation. CD127 is a hallmark memory precursor marker. during an acute immune response. We performed adoptive transfer in vivo experiments, and harvest spleens during the acute response of infection. I used this antibody in my T cell panel to identify memory precursors. I chose this product since it fits well in my 12 color panel. eBioscience/ThermoFisher was one of the few vendors first offering this marker with this particular clone.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine T cells from Spleen

Primary Incubation

1:250 (.2mg/mL Stock), 30 minutes at 4 deg or on ice

Blocking Agent

FC block cells before surface panel staining

Secondary Incubation

Results Summary

I analyzed the adoptively transferred cells during the acute response. Further analysis of the cells led to the identification of CD127+CD8+KLRG1- memory precursors.

Summary

The Good

Really nice antibody to identify memory precursors and naive CD8 T cells

The Bad

None really

The Bottom Line

This antibody clone and particular color (APC) in conjunction with KLRG1 off a different laser such as PE really leads to a nice flow plot showing the two main populations of interest.
