Obtaining purified DNA from gel electrophoresis is an important process in many molecular biology applications. In particular, many siRNA based approached such as shRNA required purified digested vector backbones for subcloning of oligo hairpin sequences. Here, I utilized a recently released NEB product for gel purification. Overall, high yields were obtained of digested purified vector that was utilized for downstream cloning procedures.

Protocol Overview

1. 4ug of digested DNA was run on a 1% agarose gel.2. The 8kb digested fragment was excised from the gel and placed into ~500ul of gel solubilization buffer3. The gel piece was dissolved for 10 mins at 45 degrees and purified as described in the kit4. 25ul of elution buffer was added to the column to elute the DNA5. Samples were measured by Nanodrop.

Tips

Warming elution buffer at 50 degrees results in better yields

Yield recovery was 70% The recovered purified DNA was utilized for downstream shRNA cloning.

Good price and easy workflow that is summarized on a tiny protocol sheet provided in the kit. Elution can be done in a tiny volume to increase the concentration of purified DNA.
