During the preparation of the libraries and performing the next generation sequencing of samples, an accurate quantification of the libraries during the pooling of different barcoded samples is needed. The Qubit™ dsDNA HS Assay Kit is highly used for the accurate quantification of the library DNA samples.

The kit is provided with HS dsDNA reagent, the dilution buffer and two standards for the calibration of the qubit machine. 200 microliter of the working solution is prepared by diluting the HS ds reagent with dilution buffer in the ratio of 1:200 for each sample. Around 190 microliters of the working solution is taken in a fresh 500 microliter tube to which 10 microliters of the diluted DNA sample is added and vortex for few seconds. Then the reading is taken using the Qubit fluorometer.
Always try to dilute the DNA sample as overloading the sample may give a false result.
Consistent results are observed using the kit and accurate quantification of the libraries was done by this kit.
