The general aim of the experiment was to generate standard curve of Nile red dye in DCM. We wanted to see the concentration of nile red within linear limit of fluorescence so as to quantify the dye in sample of interest. We selected the instrument because of its sensitivity and accuracy. We previously have noticed that this instrument can detect very low fluorescence in the order of 1 RFU or less that too with very small volume of sample (200ul).

The dye used was obtained from Sigma and was of extra purity. The solvent used was also of extra purity.

Tips

Take the blank reading in between of every reading.

Results Summary

We observed that upto around 100 ug/ml and less the curve was linear, but above that concentration the signal starts to saturate. Hence this curve can be used to quantify the dye from unknown sample if the signal intensity falls in linear range. Otherwise it is recommended to dilute the sample accordingly to bring the signal intensity within linear range.
