When substantially different levels of specific protein of interest are shown in western blots, it is extremely important to prove equal protein loading by demonstrating equal amount of proteins whose expression is relatively constant, e.g. beta-actin, alpha-tubulin or GAPDH. We used rabbit anti-beta-actin antibody to verify equal protein loading in the lysates of murine macrophages analyzed for expression of arginase-1 after cell polarization towards M1 or M2 phenotype. Rabbit anti-actin antibody from BioLegend was used to analyze the levels of beta-actin in murine macrophages expressing substantially different levels of arginase-1.

This antibody specifically recognizes beta-actin band (~40-42 kDa) and is very useful to prove equal loading when unequal expression of the protein of interest (e.g., arginase) is shown in the lysates of murine macrophages. The image shows equal protein load (similar intensity of beta-actin bands), whereas the same samples demonstrare various levels of arginase-1 expression in M2 macrophages (high), M2 macrophages re-polarized to M1 (intermediate), and M1 macrophages (almost negative).
