The aim is to investigate if macrophages phagocytose YG beads. We use a mouse macrophage cell line RAW 264.7 and Fluoresbrite® YG Microspheres 1.00µm beads. The result shows that macrophages engulf opsonized beads. So, the beads works great.

Protocol Overview

1: Culture macrophgae RAW 264.7 to 80% confluent in 2 well chamber slide. 2: Incubate YG beads in 50% mouse serum in PBS to opsonize the beads at 37C for 30min3: Add the beads to macrophages, incubate at 37C for 30 min with gently shaking.4: Wash the cell with PBS for 3 times5: Incubate cells with rat anti mouse CD68 for macophages, followed by goat anti rat alexa fluor 594.
