Pfhrp2 is a Plasmodium falciparum protein of unknown function that is produced throughout the life cycle of the parasite. In our study population, Pfhrp2 has been validated as a surrogate measure of sequestered parasite biomass, and elevated levels of Pfhrp2 are known to correlate with disease severity and adverse outcomes. We use Cellabs hrp2 ELISA kits to quantify Pfhrp2 levels in our pediatric malaria cohorts. The ELISA kits come with an easy-to-follow protocol and all of the reagents required to carry out the assay.

Protocol Overview

Samples were diluted 1:100 in PBS and subsequently plated in duplicate, along with controls/a standard curve, to the coated ELISA plate. Following a 1 hour incubation and wash steps, conjugate was added to each well. After another 1 hour incubation and wash steps, the substrate was added to each well. Each plate was left in the dark at room temperature for 15 minutes after which the stop solution in the kit was added to each well. Each plate was read ASAP at 450 nm.

Tips

Use the incubation periods to prepare for subsequent steps to reduce experiment time. Plate all samples in a separate, uncoated plate at the proper dilution so that the samples can be added to the coated plate faster using a multi-channel pipette.

Results Summary

OD values must first be obtained using a plate reader (450 nm). A standard curve can then be generated from positive control (included in the kit) or recombinant hrp2 protein dilutions. The standard curve can then be used to determine the Pfhrp2 levels in the experimental samples.

I highly recommend this ELISA kit for the quantification of Pfhrp2 in human plasma or serum samples. The kits include all reagents required for the assay including positive and negative controls. We never been disappointed with the product or the company.
