We routinely use Southern Biotech's anti-human IgG-alkaline phosphatase as the secondary antibody in an ELISA designed for detection of dsDNA. The antibody is consistent and reliable in our protocol as outlined below.

Experimental Design and Results Summary

Applications

ELISA

Sample

Human plasma, serum

Primary Incubation

Add human sample at 1:100 for 2 hours at 37C.

Blocking Agent

1% BSA in PBS

Secondary Incubation

Add anti-human IgG secondary at 1:500 for 1 hour at 37C.

Tertiary Incubation

Add phosphatase substrate solution and incubate at 37C.

Detection

Following the addition of the substrate solution, monitor the plate at determine the OD (405 nm) every 5 minutes.

Results Summary

Raw OD values must be obtained using a plate reader (405 nm). Experimental concentrations can be determined relative to a control or using a standard curve of known dsDNA concentrations.
