The VECTASTAIN ABC KIT (PK-6101) is specifically designed for immunohistochemical staining of tissues using Rabbit antibody. Here I used this kit to stain Ki67 (clone SP6, Thermo Scientific) from slides of mouse ERBB2 tumors. This kit contains biotinylated anti-Rabbit IgG that was used to bind to the primary anti-Ki67 antibody. My negative control is secondary antibody only. The protocol is simple to follow. From my years of experience,  this kit is very reliable and I always get clear staining and great results.

Mouse ERBB2 tumors

Protocol Overview

Tissue slides were prepared from mouse ERBB2 mammary tumors and stained with/without Anti-Ki67 antibody (RM-9106-R7, ready-to-use) overnight at 4 degree. After two washes with TBST for 5 minutes, slides were incubated for 30 minutes with diluted biotinylated secondary antibody (provided in the kit). Slides were then washed and incubated 30 minutes with VECTASTIN ABC reagents. After another round of wash, slides were incubated in peroxidase substrate solution until desired stain intensity develops. Rinse sections in tap water for counterstain and mount.

Tips

The incubation of secondary antiboy should not exceed 30 minutes to minimaze strong background.

Results Summary

Strong positive staining of Ki67 was achieved.
