Aim of the study was to knockdown MAPK11 in rat retina using small interfering (si) RNA approach. For this, MAPK11 siRNA, mixed with in vivo transfection reagent, were intravitreally injected into rat eyes using 33G needle. After 3 days on injection rats sere sacrificed, ratinas were harvested, protein extracts were prepared and resolved in 10% SDS-PAGE. Resolved proteins were western transferred in nitrocellulose membrane and probed for MAPK11. Antibody selection was based on previous experience with same antibody.

Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

MAPK11 antibody from Abcam showed excellent specificity towards rat proteins. There were very little or no non- specific bands. Specificity of MAPK11 Ab was also confirmed by observing specific knockdown of MAPK11 proteins.
