In the present study, tissue samples, from rats maintained at different nutritional conditions, were sampled; proteins were isolated and resolved in 10% SDS-PAGE. Resolved proteins were western transferred in nitrocellulose membrane, blocked with 5% skimmed milk and immunoprobed with Anti-ADAM 10 antibody. After secondary antibody treatment, blot was chemiluminescent developed in x-Ray film. Antibody selection was based on its specificity to rat ADAM 10 proteins.

Rat tissue whole protein lysate
Overnight at 4 0C (5% BSA in TBS with 0.1% Tween 20; dilution 1:1000)
5% Skimmed milk in TBS with 0.1% Tween 20 (for 1hr at RT)
1hr at RT (5% BSA in TBS with 0.1% Tween 20; dilution 1:5000)

ADAM10 Ab from Abcam has detected a bright band of specific ADAM 10 protein at around 90 KDa. However, there was no effect of treatment on un-cleaved ADAM 10 levels.

Some non specific bands were also visible
