This antibody was used in a flow cytometry panel in order to stain for T cells primary human PBMCs prior to identify T cell subtypes such as Th1 Th2 memory.

2µl of antibody on 1 million cells, 30 mins on ice in the dark (antibody), wash three times afterwards with 2ml of FACS buffer

Results Summary

This BV421-conjuated antibody always gives reproducible results. The OKT3 clone also works well with other colors conjugated to it We experiences better performance of this clone than the other CD3 clone from BioLegend. We used it after Ficoll gradient centrifugation. 2µl is suffiecient for 1 million cells. After incubation, be sure to wash three times with FACS buffer (HBSS, 1%BSA, 0,01% NaAzide).

Additional Notes

Be sure to block with a blockig reagent due to unselective binding of the antibodies to monocytes caused by Fc Receptors. Therefore, always compare this antibody to the corresponding Isotype.
