Protein Kinase D (PKD) is family of Ser/Thr kinases that regulate multiple biological functions including cell migration, proliferation, apoptosis, transcriptional activation, angiogenesis, and Golgi organization and vesicle trafficking. Like many other kinases, PKD functions are mediated via phosphorylation of different substrates. The goal of this project was to demonstrate if rhotekin was a bona fide substrate of PKD and whether this phosphorylation is important for regulating RhoA. In this experiment, phosphorylation of both PKD and rhotekin was tested after activating PKD with PMA.
