QIAgen mini prep kit is very useful in small scale plasmid purification. The plasmid purification is from 10 ml of bacterial starting culture. A very good plasmid yield is obtained from 10 ml bacterial culture.

The kit can be used by following the manufacturer's protocol . Briefly 10 ml of bacterial culture is pelleted down and resuspended in 250 microliters of resuspension buffer. 250 microliters of lysis buffer is added and incubated for 5 minutes. Finally, 350 microliters of neutralization buffer is added and spun at 12000 rpm for 10 minutes. The supernatent is added to the spin column and spun for 1 minute . The column is washed with PE buffer and finally eluted with buffer.
Add RNAse in the resuspension buffer
A very good quanitity of plasmid is obtained, which can be analysed using a nanospec
