Annexin V is an early apoptotic marker and is often used for detecting apoptosis in cells. Annexins are a family of calcium-dependent phospholipid-binding proteins, which bind to phosphatidylserine (PS) to identify apoptotic cells. In healthy cells, PS is predominantly located along the cytosolic side of the plasma membrane. We used this molecule to detect induced apoptosis.
Western Blot
Whole cell lysates treated with different toxic agents
Diluted 1: 500 in blocking solution, incubated for overnight at 4 degrees
5% non fat dry milk in TBST
Anti-mouse secondary antibody
None
ECl
We could determine apoptosis levels by probing for annexin V.
Reproducible results
Sometimes, it gives non specific binding to other proteins
Overall, a decent antibody for western blotting
