Protocol Overview

PureBlu Dapi reagent comes as a powder that is mixed with water to make a 100x stock. Simply dilute in PBS to a make a 1x solution that is added to the cells after the final washing step of the staining protocol. A 15 min staining with TrueBlu followed by a quick rinse in PBA will suffice for strong nuclear staining. Figure 1 shows DU145 prostate cancer cells stained with DAPI. Figure 2 shows a merge with DAPI and a FITC-labeled secondary showing cytosolic distribution of the BTF3 protein.

Tips

I used a standard protocol involving fixation with 4% paraformaldehyde, followed by permeabilization using 0.1% Triton X100 in PBS.
