ACA is an awesome centromere antibody to use, it's a human antibody, so it can be used with all other antibodies. It is a composite of cenpA, cenpB, and cenpC and works really well, but can be unspecific if used at too high of a concentration. I recommend using 1:1000-1:2000 for an hour. Here is the protocol that I used: Cells were fixed in 3.5% paraformaldehyde for 15 minutes at room temperature.3 minutes in ice cold methanol. 5 minutes in PBS+0.5% Triton X5 minutes in PBS10 minutes in PBS+3%BSA Antibody was incubated in PBS+1%BSA and coverslips incubated for 1 hour 2 - 5 minute washes in PBS Secondary antibodies from Jackson Laboratories (anti-rabbit) was incubated in PBS+1%BSA and coverslips2 - 5 minute washes in PBS Mounted coverslips using ProLong Gold
Immunofluorescence
Human cells
1:2000 in PBS+1% BSA for 1 hour at room temp
PBS+3% BSA for 10 minutes
1:1000 in PBS+1% BSA for 1 hour at room temp

Cells were co-stained with DAPI to mark the DNA (blue). ACA is stained in green. ACA is really bright and works very well! Imaging interphase cells.
