We were conducting a study of pluripotency and needed to evaluate SSEA-5 expression in a variety of cell lines. Our positive control was the H9 hES cell line from WiCell. We chose this antibody from Genetex because it was the only SSEA-5 antibody available at the time. Since our study, there have been other SSEA-5 antibodies developed, most notably from StemCell Technologies and Biolegend. This experiment tested the SSEA-5 antibody from Genetex.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

H9 human hES cells (undifferentiated), grown on a coverslip and fixed in paraformaldehyde.

Primary Incubation

Room temperature for one hour at 10 ug/ml in PBS with 0.05% Tween-20

Blocking Agent

None

Secondary Incubation

Vector Dylight 549 anti-mouse antibody (D12549) used at 5.0 ug/ml at room temperature for 45 minutes

Tertiary Incubation

Vectashield mounting medium containing DAPI counterstain

Detection

Fluorescence microscopy- Nikon microscope with SPOT camera

Results Summary

Since these H9 cells were very undifferentiated, they displayed intense staining for SSEA-5, as anticipated. This is seen in the attached photo.

The Bottom Line

The SSEA-5 antibody from Genetex works well. There are now a couple of other companies that sell SSEA-5 antibodies, so it might be worthwhile to compare them at some point. But, for the moment, this antibody works very well.
