Isolation of CD4 or CD8 T cells by antibody + magnetic beads is a routine technique used in many immunology labs and there are several kits on the market available from Miltenyi, StemCell and now eBioscience. Here, I try out the Magnisort Mouse CD4 T cell Enrichment kit from eBioscience.

Protocol Overview

1. Single cells suspension was prepared from one spleen. Spin at 2000RPM for 4 mins. 2. The pellet was resuspended with FACS buffer and adjusted to 100million cells/ml. 3. Cells were stained with 200ul of antibody per 1ml of cells at RT for 10 mins. 4. Wash cells with FACS buffer. 5. Resuspend pellet with FACS buffer and adjust cell concentration to 100 million cells/ml (the original volume). 6. Add 200ul of magnetic beads per 1ml of cells. Incubate at RT for 5 mins. 7. Bring volume to 2.5ml in a 5ml tube or 8ml if you are using a 14ml tube.8. Transfer into magnet and let sit for 5 mins.9. Pour or pipet out unbound cells depending on which magnet you are using.

Our eBioscience sales rep. assured us that the magnet we already have from StemCell will work with this kit from Ebioscience.

Results Summary

The purity obtained was around ~65-80% CD4+ T cells. This is pretty low compared to what I'm used to when using Miltenyi kit (~85-90%) and StemCell (~80-90%). Results obtained were quite variable too. Shown here are two cell separations done one after the other and I obtained purity at 66% and 75%.
