Intracellular Foxp3 were stained to evaluate T regulatory cell (Treg) population in dogs with cancer before and after treatment . We found a significantly higher percentage of Treg in dogs with cancer compared with healthy dogs. Although treatment was unable to decrease percentage of Tregs as in healthy dogs, percentage of Tregs was reduced in the dogs with cancer.

Protocol Overview

After staining with CD4 and CD25, cell were resuspended in 100ul of “cytofix/cytoperm” solution and incubated on ice for 2hrs in the dark. After washing at the end of the incubation time, cells were resuspended and incubated with fluorescent conjugated anti-Foxp3.

Tips

Incubate on ice for 2hrs in dark with "cytofix/cytoperm” solution.

Results Summary

We found a significantly higher percentage of Treg in dogs with cancer compared with healthy dogs. Although our treatment was unable to decrease the percentage of Tregs as in healthy dogs, percentage of Treg was reduced in the dogs with cancer.
