Using the Facsdiva software is very complex--a lot you have to learn and get used to. Once all the parameters and tubes are set up on the software, it is pretty easy to run your actual samples.

HEK293 cells were incubated with three different dilutions of a Thy1.1-expressing lentivirus. Cells were stained for Thy1.1 and acquired on the Fortessa.

Tips

Set up single stain controls prior to experimental sample tubes so that the brightest peak is not out of the axis.

Results Summary

This is a good machine for single or multi-color flow. The machine could be more user friendly and more intuitive. Adjusting compensation can be complicated or impossible at a few occasions. Analysis on this is poor compared to Flowjo. Gets clogged frequently.

Good machine for acquisition. Cleanup procedure is required after every person. Better than Calibur because Fortessa has more parameters. For analysis, turn to another software.
