One of the goals of my project is to carry out protein-protein interactions with components that are relevant to the hedgehog signaling. A first step in the process is to test the antibodies to determine the expression in either over-expression or endogenous levels. The goal of this experiment was to test the expression levels of Gli2 protein in NIH3T3 cells.

5% Milk in 1x TBS-T (0.1% Tween)

Donkey anti-goat IgG HRP secondary antibody for 1 hr at room temp

Tertiary Incubation

None

Detection

ECL pico for 5 min

Results Summary

The antibody detects a band at ~200 kDa (the expected size for Gli2). Based on my experience working with this antibody, it is a bit tricky working with this antibody. Sometimes you can detect the signal with ECL pico but other times, I need ECL Femto
