I bought this antibody to measure GR expression in rat brain. The specific goal was to quantify the number of GR+ neurons in the rat hippocampus and paraventricular nucleus of the hypothalamus. I chose to use this antibody because I found several papers in which the authors also used this antibody.

Primary Incubation

1:500 dilution with slices incubated at 4c overnight.

Blocking Agent

2% BSA

Secondary Incubation

Biotinylated Donkey anti-rabbit IgG (Jackson Immunoresearch) at 1/4000 dilution incubated at room temp for 2 hours

Tertiary Incubation

Vectastain ABC-kit according to manufacturer's instructions

Detection

Detected with .05% DAB enhanced by .05% Ni

Results Summary

The staining came out very clear with strong specific staining.

Additional Notes

The antibody did not work well for immunofluorescence in my hands

Summary

The Good

Easy to use with no troubleshooting required. I think that I could probably use a lower concentration than I did as well, but don't want to change my protocol now.

The Bad

I did not get very strong staining when I detected the GR primary with an Alexafluor 488-conjugated secondary

The Bottom Line

I recommend this antibody for HRP-DAB detection of GR protein in rat brain tissue.
