We were interested in examining the proliferation of the intestinal epithelial cells in healthy vs disease states. To do this, we stained for Ki67, a marker of cell proliferation.

Experimental Design and Results Summary

Immunofluorescence

Pre-fixed frozen mouse small intestine tissue section

Primary Incubation

1:100 in 1% BSA in PBS overnight at 4 degrees

Blocking Agent

5% goat serum in 1% BSA in PBS

Secondary Incubation

Goat anti-rabbit AlexaFluor488 was used at 1:200 in PBS and was incubated on the section for 1 hour at room temperature in the dark

Tertiary Incubation

Detection

Zeiss LSM 700 confocal

Results Summary

The proliferating crypt cells were clearly stained with Ki67 while the epithelial cells further up the villi did not stain. Staining was also scattered about in other lamina propria cells.
