After SDS PAGE is over, transfer the gel into transfer buffer containing box. Make sure semi-dry apparatus is working. Place the Whatman-3 filter papers on the semi-dry apparatus after soaking into the transfer buffer. Place the NC or PVDF membrane once you’re done soaking in the transfer buffer then place the SDS PAGE gel carefully without allowing to any bubbles to form. Next place the Whatman-3 filter papers. According to semi-dry user manual, supply the power and transfer will occur. Check whether the transfer has occurred by using poncueav. Once transfer is good, add the primary antibody followed by secondary antibody. After secondary incubation is over, give 3 washes with TBST and probe with ECL reagent according to GE ECL user manual. Then you should get bands without any background. We get a clean image using this procedure.

We got very good blots with out any background noise. Blots are very clean and good, publishable quality.
