Peritoneal macrophages can be easily sorted and analyzed via flow cytometry using the pan-macrophage marker - F4/80. However, to confirm and tightly control the gating, isotype controls are important. This product provides a very good negative control to analyze the F4/80 population accurately.

Protocol Overview

Standard flow cytometry protocols were used. Cells were incubated with the antibody at 1 ug/100,000 cells at 1:10 volumetric ratio for 30 mins in the dark at 4 deg C. Cells were centrifuged and resuspended in a 300-500 ul of PBS + 2% serum.

Tips

Use clean flow cytometry tubes. Accurate pipetting is important.

Results Summary

As seen in the image below, the isotype controls were slightly positive as compared to unstained cells while still low enough to mark out the F4/80+ sample. This clearly proves that isotype controls are important to setup the gates for the F4/80 analysis and sorting.
